Phase 2a study will evaluate the safety and biological activity of leronlimab in patients over the age of 50 with early-stage, biomarker-confirmed Alzheimer’s disease

VANCOUVER, Washington, June 11, 2026 (GLOBE NEWSWIRE) -- CytoDyn Inc. (OTCQB: CYDY) ("CytoDyn" or the "Company"), a clinical-stage oncology company advancing leronlimab, a first-in-class humanized monoclonal antibody targeting the CCR5 receptor with therapeutic potential across multiple therapeutic indications, today announced that the first patient has been dosed in a Phase 2a clinical study evaluating leronlimab in patients with Alzheimer’s disease, in collaboration with Weill Cornell Medicine.

The study, known as SALIENT-AD (Safety Assessment of Leronlimab and Its Effect on Neuroinflammation Targets in Alzheimer’s Disease), is a Phase 2a, open-label, proof-of-concept study designed to evaluate the safety and biological activity of leronlimab in approximately 10 to 20 patients over the age of 50 with early-stage, biomarker-confirmed Alzheimer’s disease. Patients will receive weekly subcutaneous injections of leronlimab over a 12-week treatment period. The primary endpoint will assess changes in brain inflammation and microglial activation using advanced PET imaging techniques. Secondary endpoints include safety and tolerability, cognitive assessments, blood-based biomarkers of inflammation and neurodegeneration, and measures of blood-brain barrier integrity using MRI. The study will be led by Tracy Butler, MD, Associate Professor of Neurology in Radiology and Psychiatry and Medical Director of the Brain Health Imaging Institute at Weill Cornell Medicine.

“This study is designed to evaluate whether modulation of CCR5 can meaningfully impact neuroinflammatory processes that are increasingly recognized as central to Alzheimer’s disease,” said Dr. Butler. “We are leveraging advanced imaging and biomarker tools to better understand how targeting immune signaling pathways may influence disease biology.”

“While CytoDyn remains focused on developing leronlimab in oncology, and we are especially encouraged by the early readouts from our CLOVER Phase 2 study in patients with mCRC, the collaboration with Weill Cornell Medicine in the SALIENT-AD study represents an opportunity to explore potential patient benefit in an indication that represents a significant unmet need,” said Jacob Lalezari, M.D., chief executive officer, CytoDyn.

Alzheimer’s disease is a complex and multifactorial neurodegenerative condition characterized not only by amyloid-beta plaques and tau tangles, but also by chronic neuroinflammation, microglial activation, and disruption of the blood-brain barrier. Emerging research suggests that CCR5, a chemokine receptor expressed on immune cells, may play a central role in regulating these processes.

Preclinical and translational research suggests that CCR5 plays a role in synaptic plasticity, memory formation, and microglial signaling, and may influence processes such as autophagy, vascular health, and blood-brain barrier stability. By blocking CCR5, leronlimab may help reduce maladaptive immune signaling and support neuronal resilience.

About CytoDyn
CytoDyn is a clinical-stage oncology company dedicated to advancing leronlimab, a first-in-class humanized monoclonal antibody that targets the CCR5 receptor, a key regulator of immune function implicated in cancer, infectious diseases, and autoimmune disorders.

AUSTIN, Texas & VANCOUVER, Wash.--(BUSINESS WIRE)-- Natera, Inc. (NASDAQ: NTRA), a global leader in cell-free DNA and precision medicine, and CytoDyn Inc. (OTCQB: CYDY) (“CytoDyn”), a clinical-stage oncology company advancing leronlimab, a first-in-class humanized monoclonal antibody targeting the CCR5 receptor with therapeutic potential across multiple indications, today announced a strategic collaboration to evaluate circulating tumor DNA (ctDNA) dynamics and generate real-world molecular insights to support CytoDyn’s metastatic colorectal cancer (mCRC) development program.

Under the agreement, Natera will assess CytoDyn clinical trial samples from the CLOVER Phase 2 study (ClinicalTrials.gov Identifier: NCT06699836) in patients with mCRC. SignateraTM, Natera’s personalized assay for the detection of molecular residual disease (MRD), will be used to evaluate ctDNA dynamics and molecular response patterns associated with leronlimab treatment.

Natera will also provide customized real-world data (RWD) analyses leveraging its proprietary oncology database, which is the largest multi-timepoint early- and late-stage oncology dataset with more than 2 million plasma timepoints and enriched clinical and imaging records. By integrating molecular response data from its MRD testing platform with curated electronic medical record (EMR) data, this multimodal dataset enables analyses of patient populations, treatment patterns, ctDNA response rates, and response dynamics across diverse clinical settings. Together, these capabilities are expected to generate insights into molecular response and disease progression that may help inform future clinical development of leronlimab, including clinical trial design, biomarker-driven patient selection strategies, and broader translational research efforts.

The collaboration builds on CytoDyn’s growing oncology program and follows completion of enrollment in the CLOVER study, which is evaluating leronlimab in combination with trifluridine/tipiracil (TAS-102) and bevacizumab in patients with previously treated mCRC. The collaboration is also expected to complement ongoing translational and biomarker analyses from the study aimed at further characterizing treatment response and informing future development strategies.

“Signatera has become an increasingly important tool in precision oncology and clinical development,” said Jacob Lalezari, M.D., chief executive officer, CytoDyn. “Through this collaboration, we expect to gain valuable insights into ctDNA response kinetics and disease progression that may help guide future development strategies for leronlimab in colorectal cancer and potentially other solid tumor indications.”

“We are pleased to partner with CytoDyn and provide their team with insights derived from one of the largest and most comprehensive real-world molecular oncology data platforms,” said Matt Love, vice president, biopharma data & AI partnering, Natera. “Our platform enables biopharma partners to better understand disease biology, treatment response, and patient outcomes, helping inform key development decisions throughout the drug development lifecycle.”

Program provides access to leronlimab for patients with limited treatment options while supporting ongoing clinical development

VANCOUVER, Washington, April 27, 2026 (GLOBE NEWSWIRE) -- CytoDyn Inc. (OTCQB: CYDY) ("CytoDyn" or the "Company"), a clinical-stage oncology company advancing leronlimab, a first-in-class humanized monoclonal antibody targeting the CCR5 receptor with therapeutic potential across multiple indications, including metastatic triple-negative breast cancer (“mTNBC”) and colorectal cancer (“mCRC”), today announced the successful enrollment and initial dosing of the first participant in its Expanded Access Program (EAP) for patients with triple-negative breast cancer (TNBC).

The EAP is designed to provide eligible patients access to leronlimab outside of a clinical study setting. The program is intended for patients who have exhausted available treatment options and are not eligible for ongoing or planned clinical studies, in accordance with U.S. Food and Drug Administration (FDA) guidelines.

“Dosing the first patient in our EAP marks an important step in making leronlimab available to individuals with urgent unmet medical needs, while also advancing our understanding of CCR5 biology in the treatment of aggressive cancers,” said Jacob Lalezari, M.D., CEO of CytoDyn. “Data generated through this program may further inform how CCR5 inhibition influences the tumor microenvironment, including its potential role in modulating PD-L1 expression and supporting combination approaches with immune checkpoint inhibitors.”

“For patients with advanced triple-negative breast cancer who have exhausted standard treatment options, expanded access programs can provide additional avenues for care,” said Namita Chittoria, M.D., Assistant Professor at the Huntsman Cancer Institute and the University of Utah, and a member of CytoDyn’s Scientific Advisory Board. “Leronlimab is an investigational therapy being evaluated in this difficult-to-treat setting, and access outside of a clinical study may offer a meaningful option for select patients - both as an additional treatment opportunity and as a way to preserve valuable time with family. Beyond individual use, these programs also inform our understanding of emerging therapies.”

In addition to providing compassionate access, the EAP is expected to generate real-world insights into the biological activity of leronlimab in heavily pretreated patients. Recent data, presented at the American Association for Cancer Research (AACR) Annual Meeting 2026, highlight the potential role of CCR5 inhibition in modulating the tumor microenvironment in metastatic triple-negative breast cancer, with observed associations in PD-L1 expression and broader immune signaling pathways. Together, these findings provide a scientific foundation for the EAP and support continued exploration of leronlimab as a strategy to enhance responses to immune checkpoint inhibitor (ICI) therapies.

To support execution of the program, the Company has engaged With Every Patient (WEP Clinical) as the clinical research organization to support program execution, including patient identification, site coordination, and regulatory compliance. The EAP is now open for physician referrals, and CytoDyn expects to expand participation as additional sites are activated. The program will operate under applicable U.S. Food and Drug Administration (FDA) guidelines, and additional information for physicians and eligible patients, including referral details, is available on the Company’s website at www.cytodyn.com.
